Baldur's Gate 3 will offer the quest to find the issue with the mail delivery almost immediately as you enter Act 3 of the game.
However, it is a bit mysterious since the quest marker is ambiguous and it stays on the post office even after you've exhausted the dialogue with Postmaster Danzo.
The solution is that you need someone who can speak with animals, be it through the use of a potion or spell.
Once you cast Speak With Animals, go to the upper floor and speak with the pigeon sitting on the boxes. The pigeon will direct you to your next step of the quest - you need to go west and find out what is killing the pigeon fleet, which results in their inability to deliver the mail.
Use a character with high Strength or someone who can enhance their jumping through spells or potions. Alternatively, you can use someone who can fly. Go to the roof of the temple across the street with that character and you will find the culprit - a cat with wings.
If Speak With Animals is active on your characters, you will be able to ask if she's the one killing the pigeons, which she will deny while sitting on a pile of pigeon feathers and a bundle of letters. At that point, you can pass one of several speech checks to get her to leave the pigeons alone. Failing the checks will result in her teleporting and shooting a fireball at you.
To make sure you don't get blown up, you may want to approach her with Gale. They will recognise each other as Tara the tressym is his pet and assistant, which will cause her to give up the letters without any hassle.
Once that's done, loot the letters and you can open them if you wish. This will reveal the Postmaster is dealing with the Zhentarim, which you can use to extort more money from him.
