If following the main quest or casually browsing the open maps of Nioh 3 brought you to the Hamamatsu Hill region, and naturally, you want to clear it out before proceeding further, you'll most certainly come across an enemy base guarded by the door that seemed to be locked.

Do Not Look For the Key at the Usual Places
Almost all the keys in other regions that keep such places locked are either located behind the Crucible spikes, inside enemy bases, or behind side missions that take place inside the same area.
The key that unlocks this door is indeed tied to a side quest, but you'll need to take several steps before being able to get to the part where you'll be given the key.
Two Side Quests – One Key
The person who gives you the key for the base in question is Ii Naotora. This formidable warrior can be met inside the other region called Iinoya, located farthest to the north. Here, you'll find the place called Ryotan Temple, and this lady standing in front of it, waiting for your help to clear the castle of yokai.
Once you finish this quest, she will withdraw inside Hamamatsu Castle Keep, and a new side mission called 'Ii Memento' will unlock inside the 'Progress' menu. Prioritise this mission, fast travel to the keep, and talk to Ii Naotora. She will inform you about the yokai occupying the temple near Hamamatsu Castle and give you the key to it.
Once you get the key, fast travel to the Mikatagahara Junction shrine and make your way to the locked door. Unlock it and deal with five yokai, but be aware that the final test in the form of a boss awaits you in front of the tower. Beat the boss and return to Ii Naotora. Speak to her and finish the mission, and this is how you can unlock the door to this mysterious enemy base.
