After the update 1.000.003, the game has been patched, and now every slayer can be chosen at the beginning of the first playthrough. However, you will not be able to switch slayers midgame because once you start a playthrough with one slayer, you will have to finish it. Choosing a new slayer will start a new game.

How to unlock all slayers in Dead Island 2
The good news is that there are six playable slayers in single-player mode, and the bad news is that you will have to finish the game with one of the two slayers first to gain access to the other four. Probably the most common first choice will be Jacob, a charismatic British actor who has a really specific personality perfectly fit for the zompocalyptic Hell-A.
Amy, a young woman with a disturbed personality and a running prosthetic leg, will be the second slayer you can select at the start of the game. Although she won't be as tough as Jakob, she will be extremely quick and agile, and she will be able to outrun and blindside any zombie.
After finishing the game, Dani, Carla, Ryan, and Bruno will become available. Each of those characters will have a different set of pros and cons, and it is up to you to choose which one of them fits your playstyle the best.
If you were hoping that a different slayer would have an effect on the plot and that it would change if you played with someone else, we're afraid we must disappoint you. No matter which slayer you play the game with, almost everything stays the same.
